Akash Deep made an immediate impression on debut, dismissing England's top three in a stunning new ball burst, before Bairstow and Stokes both fell to spin to leave England 112-5 at lunch.
The afternoon belonged to Root though, who knuckled down and cleverly crafted one of the best centuries of his glittering Test career in tricky conditions.
Ben Foakes played brilliantly at the other end for his 47, while Tom Hartley and Ollie Robinson also offered important resistance.
England finished the day on 302-7, with Root unbeaten on 106*.
